چکیده

Focus-engineered coherent anti-Stokes Raman scattering (FE-CARS) microscopy is used to visualize microscopic samples with differential imaging contrast in both longitudinal and lateral directions. Multidimensional contrast is achieved by applying a one-dimensional p-phase step in the transverse beam profile of the Stokes beam. The observed longitudinal differential contrast results from fortuitous phasematching at axial interfaces and is highly sensitive to the resonant part of the third-order susceptibility. Based on phase-sensitive recordings of the vibrational spectrum of dimethyl sulfoxide in the CH3stretching range, it is shown that FE-CARS uniquely probes the spectral phase of the Raman bands of the sample. ! 2008 Elsevier B.V. Laser-scanning coherent anti-Stokes Raman scattering microscopy and applications to cell biology.

Laser-scanning coherent anti-Stokes Raman scattering (CARS) microscopy with fast data acquisition and high sensitivity has been developed for vibrational imaging of live cells. Dual/differential coherent anti-Stokes Raman scattering module for multiphoton microscopes with a femtosecond Ti:sapphire oscillator.

In the last decade, coherent anti-Stokes Raman scattering (CARS) microscopy has emerged as a powerful multiphoton imaging technique offering label-free chemical sensitivity and high three-dimensional resolution. However, its widespread application in the life sciences has been hampered by the use of costly pulsed lasers, the existence of a nonresonant background requiring involved technical sol...

Hyperspectral and differential CARS microscopy for quantitative chemical imaging in human adipocytes.

In this work, we demonstrate the applicability of coherent anti-Stokes Raman scattering (CARS) micro-spectroscopy for quantitative chemical imaging of saturated and unsaturated lipids in human stem-cell derived adipocytes. We compare dual-frequency/differential CARS (D-CARS), which enables rapid imaging and simple data analysis, with broadband hyperspectral CARS microscopy analyzed using an uns...

Chemically-selective imaging of brain structures with CARS microscopy.

We demonstrate the use of coherent anti-Stokes Raman scattering (CARS) microscopy to image brain structure and pathology ex vivo. Although non-invasive clinical brain imaging with CT, MRI and PET has transformed the diagnosis of neurologic disease, definitive pre-operative distinction of neoplastic and benign pathologies remains elusive. Definitive diagnosis still requires brain biopsy in a sig...
